The most important issue is not dosage. It is knowing what the product contains.
Diosgenin is a naturally occurring steroidal sapogenin found in certain plants. Its steroid-like ring structure makes it useful as a chemical starting material, but that does not mean the human body converts an oral diosgenin supplement into testosterone or an anabolic drug.
“6-Keto diosgenin” is used as a supplement-market term rather than a consistently verified ingredient identity. One seller may mean a specific derivative; another may use it loosely for a product related to laxogenin or 5α-hydroxy-laxogenin.
That ambiguity makes broad claims especially unreliable. A study on one compound cannot automatically validate another compound with a similar name.
Laxogenin is a plant steroidal compound. 5α-hydroxy-laxogenin is a different derivative that is often marketed as plant-derived or “natural.” Newer research describes 5α-hydroxy-laxogenin as synthetic rather than a naturally occurring plant compound, and the FDA has challenged its use in U.S. dietary supplements.
These are ester names. In prescription anabolic steroids, an ester can alter release characteristics after injection. That does not prove an oral 6-keto diosgenin ester has the same absorption, half-life, conversion, or anabolic effect.
Unless a manufacturer provides verified identity testing and human pharmacokinetic data, names such as “cypionate” or “decanoate” are chemistry labels—not evidence of longer action or better muscle growth.
The most frequently cited evidence is a 1976 Russian rat study by Syrov and Kurmukov.
Researchers tested several 6-keto derivatives of natural sapogenins. One derivative increased body weight and the weight or protein content of several organs and a leg muscle in rats. The paper also reported no androgenic activity in that model.[1]
That finding is not enough to show that a modern capsule works in people.
Calling 6-keto diosgenin “proven anabolic” turns a narrow animal observation into a conclusion the study did not test.
Because supplement listings often blur 6-keto diosgenin with laxogenin-related compounds, newer 5α-hydroxy-laxogenin research is relevant—but it still cannot be treated as direct evidence for every product using the 6-keto name.
A 2022 cell-based study found that 5α-hydroxy-laxogenin could bind to and activate the androgen receptor in an in-vitro system.[2]
This is mechanistic evidence. It shows biological interaction under laboratory conditions, not muscle growth, testosterone elevation, or safety in a person taking capsules.
A 2025 study then tested 5α-hydroxy-laxogenin in orchiectomized rats for two weeks. It found no androgenic effect in prostate, seminal vesicle, or penile tissue and no anabolic effect in the levator ani or hindlimb muscles. At the highest dose, some tissues showed greater atrophy rather than growth.[3]
The researchers concluded that the in-vitro receptor signal did not translate into an anabolic or androgenic effect in that animal model.
This still is not a human oral trial, but it directly weakens the common claim that receptor activity automatically means a useful anabolic effect.
A 2026 metabolism paper identified several 5α-hydroxy-laxogenin metabolites in cell and rat models. It noted that the compound was not prohibited by WADA at the time of publication because available data did not justify inclusion—but it also emphasized that human performance and health-risk data remain insufficient.[4]
“Not currently prohibited” is not the same as safe, legal to sell as a supplement, proven effective, or guaranteed not to create a testing problem through contamination.
There is no reliable controlled human evidence that oral 6-keto diosgenin increases testosterone, luteinizing hormone, or free testosterone.
This claim is often built from one of three errors:
A product can have a steroid-shaped molecule without acting like testosterone in the human body. Chemical resemblance is a reason to study a compound—not proof of an outcome.
No controlled oral human trial has shown that 6-keto diosgenin increases lean mass, muscle thickness, one-repetition maximum strength, training volume, or recovery.
That distinction matters. A bodybuilding supplement should not earn a recommendation based only on:
For muscle and strength, training progression, sufficient dietary protein, energy intake matched to the goal, sleep, and well-studied supplements such as creatine have materially stronger evidence.
A responsible answer is not that 6-keto diosgenin has “no side effects.” The responsible answer is that the side-effect profile is not adequately characterized in humans.
Key unknowns include:
People who are pregnant or breastfeeding, under 18, managing a hormone-sensitive condition, taking medication, preparing for surgery, or dealing with liver, kidney, cardiovascular, or endocrine issues should not experiment with a poorly characterized ingredient without medical guidance.
The FDA has specifically stated that 5α-hydroxy-laxogenin does not qualify as a dietary ingredient, is not generally recognized as safe for its marketed use, and makes products containing it adulterated under U.S. law.[5][6]
The agency’s current ingredient directory continues to list 5α-hydroxy-laxogenin and several aliases, including decanoate and undecanoate forms, alongside its enforcement history.[7]
Two cautions are necessary:
Vagueness does not make the product safer. It makes it harder to evaluate.
Do not rely on a seller’s statement that 6-keto diosgenin or laxogenin “will not show on a drug test.”
A 2026 analytical study reported that 5α-hydroxy-laxogenin itself was not prohibited by WADA at that time, but the authors also described it as an illegal supplement ingredient and characterized metabolites that could be used for screening.[4]
USADA has warned that products listing laxogenin have frequently overlapped with high-risk bodybuilding supplements, including products that listed or tested positive for prohibited substances.[8]
Competitive athletes remain responsible for what enters their body. The lower-risk approach is to avoid obscure muscle-building products and use only batch-certified supplements after checking the current rules with the applicable anti-doping organization.
No evidence-based human dose exists for 6-keto diosgenin.
Amounts copied from product labels, bodybuilding forums, or unrelated diosgenin studies are not validated dosing guidance. The same applies to cycling schedules and claims that one ester should be dosed less often than another.
Without human pharmacokinetic, efficacy, and safety studies, a precise dose would create false confidence.
6-Keto diosgenin does not currently meet a reasonable evidence threshold for a muscle-building recommendation.
The best-known support is an old rat paper on related sapogenin derivatives. Newer work on the commonly associated compound 5α-hydroxy-laxogenin found receptor activity in cells but no anabolic or androgenic effect in a short rat model. No controlled human trial establishes muscle gain, strength, testosterone elevation, a useful dose, or long-term safety.
Recommendation: skip it. The uncertain identity, lack of human efficacy data, and regulatory concerns surrounding 5α-hydroxy-laxogenin make the risk-to-evidence ratio poor.
